Efficiency of the Establishment of Sheep Embryonic Stem Cell Lines from Whole Blastocysts and Isolated Inner Cell Masses

Abstract

|
Eighteen embryoinc stem(ES) cell lines (26%, 18/70) were established from 70 sheep whole blastocysts and isolated inner cell masses(ICM). The cells could be maintained undifferentiated on mouse fibroblast feed layer. The efficiency of isolation
of
ES-like colonies and established ES cell lines were higher (p<0.05, by X©÷analysis) from isolated ICMs than from whole blastocysts. ES cell lines spontaneously differentiated into embryoid bodies in suspension culture, endoderm-like,
muscular-like,
neuron-like cells, lipid vesicle and stained positive for alkaline phosphatase activity. We are currently attempting to evaluate whether these cultures represent primary lineages which would be of particular relevance to developmental and
transgenic
studies.
